I read that you haven’t changed anything, but often that message is misleading and means that either the folder or the access database is lacking write permissions.
Despite what it says it is probably nothing the do with the query per-se as the update query that you mention is not one that is likely to fail with that error as it is a simple update.
If you right click on the folder and the database from Windows Explorer you may get a security tab, if you do check that write permissions on both the folder and the mdb are set.
If you don’t get a security tab, then goto Tools-Folder Options->View->Use Simple File Sharing and untick it. Then do the above paragraph.
There is no obvious reason why permissions would have changed from what you have said but.....
