Suppose if I have a website and configured it as www.mydomain.com:8182 which is the address of Computer 1 in my local network. Now I have changed and now www.mydomain.com:8182 points to Computer 2. Users will be directed to Computer 2 because of new settings. I want to know that how it internally works within my local network? any hepl will be appreciated.

Ports are usually directed in your Router (it being a Soft- or Hardware Router). In the Router interface you have WAN adresses listed as well as LAN adresses. So if a request comes in on the WAN address 164.0.0.1:8182 it hits your router, the router uses that routing table to decide where to pass the request to i.e. 192.168.1.9:8182 (it can be a totally different port for that matter). So if you change the IP/Name of the LAN computer you direct it to a different computer, they are furter divided in udp and tcp requests.
