If you can only write one comment on a post can not be reached to initiate a discussion on the content of the post. You can put the user can write x comments per hour.

If you disable 'Anti-Spam Cookies' then you can have people comment multiple times.

The 'Anti-Spam IP Blocking' only checks the last comment IP, so if you keep this enabled it will stop your blog being flooded with spam while still allowing someone to reply to a comment from someone else.
